Description
Tth Polymerase, 5 U/µl is a recombinant thermostable DNA polymerase derived from Thermus thermophilus HB8 and expressed in E. coli. In addition to its DNA-dependent DNA polymerase activity, the enzyme exhibits RNA-dependent DNA polymerase activity in the presence of Mn²⁺ ions, enabling reverse transcription and PCR amplification with a single enzyme.
Unlike conventional reverse transcriptases, Tth Polymerase combines reverse transcription and PCR amplification in one workflow, making it particularly suitable for one-step RT-PCR applications. The enzyme lacks RNase H activity and can efficiently reverse transcribe and amplify RNA targets up to 2–3 kb in length.
Tth Polymerase accepts a wide range of modified nucleotides and can therefore be used for the incorporation of labeled dNTPs, including biotin-, digoxigenin-, and fluorescein-modified nucleotides. This makes the enzyme an excellent choice for probe generation, nucleic acid labeling, and molecular detection assays.
The supplied reagent system includes dedicated buffers for PCR, reverse transcription, and one-step RT-PCR applications, as well as MnCl₂ required for RNA-dependent DNA polymerase activity.
For applications requiring maximum convenience and reduced contamination risk, one-step RT-PCR can be performed using the supplied RT-PCR buffer. For downstream applications where higher sequence fidelity is required, a two-step RT-PCR workflow is recommended, as the presence of Mn²⁺ ions may increase the polymerase error rate.
