Description
AptaHotTaq DNA Polymerase, 5 U/µl is an advanced HotStart PCR enzyme consisting of recombinant Taq DNA polymerase complexed with synthetic DNA aptamers. During PCR set-up at ambient temperature, the aptamers inhibit polymerase activity, preventing non-specific amplification and primer-dimer formation. At temperatures above 50 °C, the aptamers reversibly dissociate from the enzyme, automatically activating the polymerase during standard PCR cycling conditions.
The enzyme efficiently amplifies DNA fragments up to 5 kb and is ideally suited for demanding PCR applications including complex genomic DNA, cDNA templates, low copy number targets, multiplex PCR, and real-time PCR.
AptaHotTaq DNA Polymerase possesses a robust 5’→3’ polymerase activity and generates PCR products with 3’-A overhangs, making it compatible with standard TA cloning workflows. The optimized aptamer-based HotStart technology provides excellent specificity, clean amplification results, and convenient room temperature reaction set-up without the use of antibodies.
For maximum flexibility in assay optimization, the polymerase is supplied with three different 10x reaction buffers:
- Incomplete NH4 buffer (without MgCl2)
- Complete NH4 buffer
- Complete KCl buffer
Additional MgCl2 solution is included for individual optimization of magnesium concentration according to specific assay requirements.
AptaHotTaq DNA Polymerase is also available as a high-concentration variant (30 U/µl) for diagnostic kit manufacturing and specialized workflow applications.
