Description

SmartHotTaq DNA Polymerase, 5 U/µl is a genetically modified recombinant Taq DNA polymerase combined with anti-Taq monoclonal antibodies for reliable HotStart PCR performance. The antibody-mediated inhibition prevents polymerase activity during reaction set-up at room temperature, minimizing non-specific amplification and primer-dimer formation. Full enzyme activity is automatically restored during the initial denaturation step above 70 °C.

The enzyme efficiently amplifies DNA fragments up to 5 kb and is specifically designed for challenging PCR applications, including highly multiplexed assays and samples containing PCR inhibitors. Its optimized formulation provides enhanced robustness, high specificity, and reliable amplification even under demanding conditions.

SmartHotTaq DNA Polymerase possesses a robust 5’→3’ polymerase activity and generates PCR products with 3’-A overhangs, making it compatible with standard TA cloning workflows.

For maximum flexibility in assay optimization, the polymerase is supplied with three different 10x reaction buffers:

  • Incomplete NH4 buffer (without MgCl2)
  • Complete NH4 buffer
  • Complete KCl buffer

Additional MgCl2 solution is included for individual optimization of magnesium concentration according to specific assay requirements.

For applications that do not require HotStart technology, SmartHotTaq DNA Polymerase is also available as the antibody-free variant SmartTaq DNA Polymerase.
