Description
SD Polymerase HotStart, 10 U/µl, is a novel artificial thermostable DNA polymerase engineered for strong strand displacement activity and high-performance DNA amplification. The enzyme is stable up to 93 °C and combines robust PCR capability with isothermal amplification strategies in a single system.
The enzyme possesses both 5’→3’ polymerase activity and 5’→3’ strand displacement activity, while lacking exonuclease activity. It generates PCR products with 3’-A overhangs, making it compatible with standard TA cloning workflows.
In the HotStart format, SD Polymerase is optimally blocked by antibodies, ensuring complete enzyme inactivation during reaction set-up at room temperature. Activity is fully restored upon initial heating, enabling highly specific and controlled PCR performance with minimal non-specific amplification.
SD Polymerase HotStart is particularly efficient for GC-rich and structurally complex templates and enables reliable amplification of challenging targets. It supports long-range PCR of up to 30 kb and is suitable for a wide range of applications including standard PCR, library preparation, whole genome amplification (WGA), strand displacement–based assays, and isothermal amplification workflows such as PCDR.
For LAMP applications, the non-HotStart SD Polymerase (50 U/µl) is recommended, as optimal performance in isothermal formats is achieved without antibody-based inhibition.
The enzyme is fully compatible with workflows requiring both PCR and isothermal amplification, and initial denaturation steps can further improve template accessibility and reaction efficiency.
SD Polymerase HotStart is part of a versatile enzyme family also available in antibody-free and high-concentration formats (50 U/µl) for large-scale applications.
