Description
Oligo(dT) Primer is a highly purified oligonucleotide consisting of 20 deoxythymidine residues [Oligo(dT)20], specifically designed for first-strand cDNA synthesis from polyadenylated mRNA templates. By annealing to the poly(A) tail of eukaryotic messenger RNA, the primer enables selective reverse transcription of mature mRNA and supports the generation of full-length cDNA for downstream molecular biology applications.
The primer provides highly efficient initiation of reverse transcription reactions and is particularly suitable for gene expression analysis, RT-PCR, RT-qPCR, cDNA library construction, next-generation sequencing (NGS), and other RNA-based workflows requiring specific conversion of mRNA into cDNA.
Oligo(dT) Primer is HPLC purified to ensure maximum purity, reproducibility, and performance in sensitive molecular biology applications. The product is supplied in lyophilized form for convenient storage and long-term stability and can be easily reconstituted with the included nuclease-free water.
For improved transcript coverage and enhanced sensitivity, especially when working with partially degraded RNA or transcripts containing strong secondary structures, the use of a 1:1 mixture of Oligo(dT) Primer and Random Hexamer Primer is recommended. This combined priming strategy enables efficient reverse transcription of both full-length mRNA molecules and additional transcript regions that may not be efficiently accessed by Oligo(dT) primers alone.
