Restriction endonucleases are used in biotechnology for gene cloning, to cut DNA in smaller strands to study similarities and differences in fragment length among individuals like in RFLP (Restriction Fragment Length Polymorphism). BIORONs restriction enzymes show high purity and activity.
Not I can be heat-inactivated with 20 min at 65 °C and shows 75 – 100% activity in BIORON buffer H and 50 % activity in BIORON buffer K (supplied with the enzyme). Please note that activity is blocked by CpG methylation. Super coiled plasmids may require up to 5-fold more Not I for complete digestion than linear DNAs.
