Cutting sequence: G↓ANTC
Isoschizomere: N/A
Source: An E. coli that carries the cloned Hinf I gene from Haemophilus influenzae Rf.
Buffer supplied: 10x H.
Substrate for unit definition: λ DNA.
Reaction conditions: 100 mM NaCl, 50 mM Tris-HCl (pH 7.9 at 25oC) , 10 mM MgCl2, 1 mM dithiothreitol, 100 μg/ml bovine serum albumin and DNA. Incubate at 37°C.
Storage buffer: 50 mM KCl, 10mM Tris-HCl (PH 7.4 at 25°C), 0.1 mM EDTA, 1mM dithiothreitol, 200 μg/ml bovine serum albumin and 50 % glycerol. Store at -20°C
Absence of contaminants: Thirty units of Hinf I do not produce any unspecific cleavage productsafter 16 hrs incubation with 1 μg of λ DNA at 37°C. After ten-fold overdigestion with Hinf I, greater than 90% of the DNA fragments can be ligated and recut with this enzyme.
Heat inactivation: 80°C for 20 minutes.
