Description:
Pfu DNA polymerase is a thermostable enzyme isolated from Pyrococcus furiousus. The enzyme replicates DNA at 75oC, catalyzing the polymerisation of nucleotides into duplex DNA in the 5´--> 3´direction. Pfu DNA Polymerase possesses 3' --> 5' exonuclease (proofreading) activity. Nucleotide misinsertions that may occur during polymerization are rapidly excised by the proofreading activity of the polymerase. Pfu DNA Polymerase is recommended for use in PCR and primer extension reactions that require high fidelity synthesis. Pfu DNA Polymerase - generated PCR fragments are blunt - ended.


Concentration :
5000 units/ml

Storage buffer:
50 mM TrisHCl (pH 8,2), 0.1 mM EDTA, 1mM DTT, 0,1% Nonidet P40, 0,1% Tween-20, 50% glycerol.

Supplied Reaction Buffers (10x):  

  1. Dilution buffer: 50 mM Tris-HCl (pH 8,2), 0.1 mM EDTA, 1mM DTT, 0,1% Nonidet P40, 0,1% Tween-20, 50% glycerol.
  2. Complete: 200 mM Tris-HCl (pH 8.8 at 25° C), 100 mM KCl, 100 mM (NH4)2SO4, 20 mM MgSO41,0% Triton X-100
  3. Incomplete:  200 mM Tris-HCl (pH 8.8 at 25° C), 100 mM KCl, 100 mM (NH4)2SO4, 1,0% Triton X-100; supplied with 1 ml of 25 mM  MgSO4 separately packed.
