Introduction
The Ron’s family of products is an innovative system that radically simplifies extraction and purification of nucleic acids from a variety of sources. Ron’s Plasmid Midi Kits employ a modified alkaline/SDS lysis procedure to prepare plasmids from bacterial culture. There is no need for organic extraction methods like phenol or chloroform. Yields vary according to plasmid copy number, E. coli strain and conditions of growth, but 30 ml of overnight culture with high copy-number plasmids in LB medium typically yield 20 – 100 µg highly pure plasmid DNA. For more Plasmid DNA we recommend the use of Ron’s Plasmid Maxi Kits, that typically yield 100 – 500 µg of high-copy plasmid DNA from 30 – 150 ml culture volume.
Plasmid-DNA isolated with the Ron’s Plasmid Midi Kits is suitable for automated fluorescent DNA sequencing, cloning, restriction endonuclease digestion, in-vitro translations and other applications.
Principle
The Ron’s Plasmid Midi Kits combine the power of XChange technology with the time-tested consistency of alkaline-SDS lysis of bacterial cells to deliver high quality DNA. Plasmid DNA binds effectively to the anion-exchange resin of the XChange Midi column and can easily be purified from contaminants and enzyme inhibitors. Purified plasmid DNA is simply eluted from the column in Elution buffer. The XChange Midi column binding capacity is 100 µg. The protocols are suitable for purifying most plasmids ranging from 3 - > 10 kb, cosmids from 10 – 50 kb, and very large constructs (P1 constructs, BACs, PACs) up to 300 kb.
